The BIOSYNEX AMPLIQUICK® Fecal Pretreatment kit is a manual kit for pretreating fecal samples to screen for intestinal parasites using gene amplification.

The kit comes in pre-filled vials with ready-to-use grinding beads and pretreatment buffer, designed to release nucleic acids from the biological samples. It is intended for use in molecular diagnostics only and is validated for use with the BIOSYNEX AMPLIQUICK® Helminths and Protozoans PCR kits. The kit optimizes the release of DNA from biological samples for molecular detection testing and is based on the physico-chemical lysis of the cell membrane components or the wall of parasites, cystic forms or eggs.

The kit’s lysis stages are optimized to increase the ratio of non-human DNA extracted. After centrifugation, the DNA remains suspended in the supernatant and can be extracted using a suitable kit.

Specifications of the Fecal Pre-treatment Kit

BIOSYNEX AMPLIQUICK® Fecal Pretreatment is a manual kit designed to screen for intestinal parasites through gene amplification. It comes with pre-filled vials containing ready-to-use grinding beads and pretreatment buffer to release nucleic acids from biological samples. The kit is intended to be used by laboratory staff for in vitro molecular diagnostics only and is validated for use with BIOSYNEX AMPLIQUICK® Helminths and Protozoans PCR kits. The kit optimizes the release of DNA contained in biological samples to perform molecular detection testing, which helps identify both silent and acute infections. The kit uses a physico-chemical lysis method, involving mechanical grinding and heating to 95°C to increase the ratio of non-human DNA extracted.

The pretreatment medium is specifically designed to allow DNA to separate from impurities, PCR inhibitors, and enzymes present in large quantities in feces. After centrifugation, the DNA remains suspended in the supernatant, while the sample matrix is eliminated in the pellet. This kit is an essential tool for in vitro molecular diagnostics, providing accurate and sensitive detection of intestinal parasites in fecal samples.

Components of the Fecal Pre-treatment Kit
  1. Lysis Buffer: A solution containing chaotropic salts and detergents that break down the cell walls and membranes of fecal bacteria to release genomic DNA into solution.
  2. Proteinase K: An enzyme that digests proteins and enzymes, including nucleases and proteases that could degrade the extracted DNA.
  3. Beads: Silica-coated magnetic beads that capture and isolate genomic DNA from the lysed fecal samples.
  4. Wash Buffer: A solution used to wash the magnetic beads and remove any remaining contaminants.
  5. Elution Buffer: A solution used to release the captured genomic DNA from the magnetic beads.
  6. DNase/RNase-free Water: Water that has been treated to remove contaminating DNases and RNases, which can degrade the extracted DNA.

 

The kit also includes instructions for use, as well as a protocol for the extraction of genomic DNA from fecal samples using a magnetic bead-based method.
