Introduction

The FGB gene encodes the fibrinogen β-polypeptide, a vital component in the blood clotting process. When cleaved by thrombin, fibrinogen forms fibrin, a key factor in coagulation. Studies have established a clear link between the -455A allele and higher fibrinogen levels in the general population. Homozygosity for the A allele, in conjunction with other genetic and environmental factors, may increase the risk of coronary and cerebral events.

Storage and Stability

Store the kit at -30 to -15°C upon arrival. Alternatively, it can be stored at 2 to 8°C for short-term use within one month. The kit remains active through up to 20 freeze/thaw cycles without loss of functionality. Principle of the Test: The FGB -455G>A RealFast™ Assay employs the fluorogenic 5’ nuclease assay, also known as the TaqMan® assay. Each reaction involves a gene-specific primer pair that amplifies a 104 bp fragment of the FGB gene, along with two dual-labeled, allele-specific hydrolysis probes that bind to the target sequence of the amplified fragment. During PCR’s extension phase, the Taq DNA polymerase’s 5’ – 3’ exonuclease activity cleaves the 5’-fluorescent reporter from the hybridized probe. This cleavage results in a fluorescent signal proportional to the accumulated PCR product
