Understanding the Functionality of the Detection Kit
The Imegen M-BCR-ABL1 (IMG-121) detection kit employs a combination of oligonucleotides and hydrolysis probes for the efficient amplification and quantification of M-BCR-ABL1 (p210) rearrangement. This real-time PCR tool is designed to enable lab scientists to calculate the number of copies of the rearrangement and the number of copies of the endogenous ABL gene. Furthermore, this genetic analysis allows the detection of minimal residual disease (MRD), with the sensitivity level contingent on the maximum number of detectable copies of the ABL1 reference gene.
To achieve the necessary sensitivity limit for MRD analysis that would result in a clinically significant molecular response, the process of total RNA extraction from peripheral blood or bone marrow cells needs to be optimized. Consequently, the reverse transcription protocols should be maximized to yield the greatest amount of complementary DNA (cDNA) in each sample.
The detection kit uses the standardized calibrator available at the Institute of Reference Materials and Measurements (IRMM), Belgium, catalog number ERM-AD623, for calibrating the pIMEGEN plasmid (M-BCR-ABL1:ABL1 at a copy ratio of 1:1). This plasmid is used as a standard for the quantification performed with the kit.
