MolYsis Complete5 combines sample pre-treatment and microbial DNA isolation in one kit, for background-free PCR on whole blood and other body fluids. Human cells are lysed and the released human DNA degraded before the bacterial and fungal DNA is purified. Research use only. Four parallel preparations take about 120 minutes. Protocols are provided for samples up to 1 ml and for 5 ml.

MolYsis Complete5 isolates bacterial and fungal DNA from clinical material with a large excess of human DNA, and covers the whole path from sample to purified microbial DNA.

Principle

A chaotropic buffer lyses the human cells in the sample while microbial cells stay intact. The human DNA released, including DNA from dead cells, is degraded by the chaotrope-resistant DNase MolDNase B. The microbial cells are then collected and their cell walls degraded with BugLysis, followed by Proteinase K digestion and spin-column purification of the microbial DNA.

Sample material

Human material: whole blood with anticoagulant, synovial fluid, pleural fluid, cerebrospinal fluid, ascites, pus, bronchoalveolar lavage, nasal douche and urine. Animal material: whole blood and cell cultures. There is a protocol for samples up to 1 ml and one for 5 ml. Frozen samples without a cryoprotectant are not suitable.

Practical points

Four parallel preparations take about 120 minutes. Besides a thermomixer and a bench-top microcentrifuge, a high-speed centrifuge for 50 ml tubes at 9,500 x g is required for the larger volume. The manual lists the tested Gram-negative and Gram-positive bacteria, fungi and protists, without giving a total count. No controls are supplied. Shelf life is 24 months; enzymes are stored frozen and buffers at room temperature. The manual warns against adding hypochlorite or acid to the preparation waste.

Regulatory status

This kit is for research use only and not for use in diagnostic procedures.

  1. Lysis Buffer: A unique lysis buffer designed to efficiently disrupt the cell membranes of bacterial and fungal cells, releasing DNA for downstream applications.
  2. Magnetic Beads: Magnetic beads are used to bind to the released DNA, allowing for easy isolation and purification of DNA from other cellular components.
  3. Wash Buffers: The kit includes optimized washing steps that effectively remove contaminants, ensuring high-quality DNA yields.
  4. Elution Buffer: A buffer used to elute the purified DNA from the magnetic beads.
  5. DNase/RNase-free Water: Water that has been treated to remove contaminating DNases and RNases, which can degrade the extracted DNA.
  6. Tubes and Tips: The kit includes tubes and tips necessary for sample preparation and DNA extraction.
