Ashkenazim Son gDNA (EGFR) (SID-0004) is human genomic DNA from the GM24385 cell line, supplied as the wild-type background for EGFR mutation testing. The mutation status and the whole genome sequencing data of the line are publicly available through the Personal Genome Project. As a wild-type control it shows whether an EGFR assay stays negative where it should, and it serves as the background into which a mutation standard is diluted to a chosen allele frequency.

Ashkenazim Son gDNA (EGFR) is the wild-type material in the EGFR series: genomic DNA from a published reference genome, without the EGFR variants that the mutation standards carry.

Two uses

As a negative control it establishes the false positive rate of an EGFR assay: any signal is by definition unwanted. As a diluent it sets the allele frequency, since a mutation standard diluted in wild-type DNA gives a series at a known mutant fraction.

Reference genome

The GM24385 line is part of the Personal Genome Project and its whole genome sequencing data and mutation status are freely accessible, so a result obtained with this DNA can be compared against a known dataset. The material is therefore also usable for validating whole genome and amplicon sequencing and PCR protocols and for calibrating fragmentation equipment.
