CLIA Adiponectin
CLIA Adiponectin is a chemiluminescent immunoassay for the quantitative determination of adiponectin in human serum on the KleeYa analyser. The measuring range is 12.5 to 800 ng/ml, the sample volume 13 µl and the assay time 15 minutes. Adiponectin is produced by adipose tissue and has antidiabetic and anti-inflammatory properties. Concentrations fall as visceral fat accumulates, and low levels are associated with type 2 diabetes, coronary atherosclerosis, hypertension and left ventricular hypertrophy.

CLIA Adiponectin measures adiponectin in serum, as part of the assessment of metabolic disorders.
Clinical background
Adiponectin is secreted by adipocytes and acts to improve insulin sensitivity and to dampen inflammation. Unlike most adipose-derived hormones its concentration falls as visceral fat increases, and low concentrations are associated with type 2 diabetes, coronary atherosclerosis, hypertension and left ventricular hypertrophy. The manufacturer also puts forward the ratio of adiponectin to leptin as an indicator of adipose tissue dysfunction: a low ratio may point to leptin resistance and a higher likelihood of insulin resistance. The companion assay for that ratio is CLIA Leptin (CL-LEP050), with a measuring range of 1 to 64 ng/ml and an assay time of 22 minutes.
Method
The assay is a chemiluminescent immunoassay in which immune complexes are separated on magnetic particles and the signal is generated as flash chemiluminescence from an acridinium ester, measured in relative light units. The measuring range is 12.5 to 800 ng/ml, the sample volume 13 µl of serum and the assay time 15 minutes.
Platform
The kit runs on the KleeYa analyser, an open platform, and includes the reagents needed with the calibrators. The control set (CL-ADPCON, 2 x 20 tests) with positive and negative control sera is ordered separately; its control limits are stated on the certificate. Reagent consumption and test counts are tracked by RFID tags and the analyser supports a connection to the laboratory information system.
