Hepatitis C Virus (HCV) PCR Kit

The TruPCR HCV Viral Load Kit quantifies hepatitis C virus RNA in serum or plasma by real-time RT-PCR, across all genotypes and subtypes. The assay is calibrated against the WHO international standard and reports in international units per millilitre. Research use only. The stated limit of detection is 20 IU/ml. A five-point standard curve and an internal control that targets a human gene are included, so extraction and amplification efficiency are checked in every sample.

The TruPCR HCV Viral Load Kit measures the hepatitis C viral load, the parameter that determines whether antiviral treatment is working. Research use only.

Clinical background

Hepatitis C virus is an RNA virus that causes chronic liver inflammation and, over years, fibrosis, cirrhosis and hepatocellular carcinoma. Direct-acting antivirals cure most infections, and the viral load is what shows whether the virus has been cleared. Because six genotypes circulate, an assay used for monitoring has to detect all of them equally.

Method

Real-time RT-PCR on a conserved region of the viral genome, with dual-channel detection: one channel for the target and one for the internal control, which targets a human gene sequence and so monitors extraction and amplification per sample. Quantification runs against a five-point standard curve calibrated to the WHO international standard, so results are reported in IU/ml. The stated limit of detection is 20 IU/ml. The kit contains the primer and probe mix, ready-to-use reverse transcription and amplification reagents and the quantitation controls.
