Mastermix 16S Basic Kit

Mastermix 16S Basic is a DNA-free 2.5x PCR master mix with MolTaq 16S polymerase, for the amplification of bacterial and fungal DNA with primers of your own choice. Research use only. The DNA-free grade is the point of the product: the manufacturer specifies an absence of signal in negative controls in at least 97 per cent of cases over up to 40 cycles. Primers are not supplied.

Mastermix 16S Basic is the DNA-free master mix for laboratories that run their own primer sets. The intended use in the datasheet is the PCR amplification of bacterial and fungal DNA using custom primers.

Why DNA-free matters

A broad-range bacterial PCR amplifies any bacterial DNA present, including the traces carried by ordinary reagents and enzymes. That background, not the assay design, sets the detection limit. This master mix is manufactured DNA-free, and the specification is that negative controls stay clean in at least 97 per cent of cases for up to 40 cycles.

What is and is not supplied

The kit contains the 2.5x concentrated master mix at 3 mM magnesium final concentration, the MolTaq 16S DNA polymerase in a non hot start version, and DNA-free PCR-grade water. It contains no primers: the primer set is chosen by the laboratory. The reaction volume is 25 µl and the cycling conditions follow the assay in use.

Controls

DNA-free water serves as the negative control. For the positive control, genomic DNA from Staphylococcus aureus or another species is used, or the separate Molzym DNA positive control (REF S-200-050). Storage is at -15 to -25 °C and the mix is stable for 24 months.

The Mastermix 16S Basic is the main component of the kit and contains all the necessary reagents for PCR amplification of bacterial 16S rRNA genes. The positive control DNA and negative control DNA are provided to verify the performance of the kit and to detect any potential contamination. Sterile water is provided to dilute the DNA controls and to adjust the final reaction volume. PCR tubes are also included in the kit.

The kit does not include the template DNA or DNA-free water, which need to be added by the user. Additionally, the kit includes a comprehensive user manual that provides detailed instructions for the preparation and use of the Molzym Mastermix 16S Dye, as well as troubleshooting tips and advice on data interpretation.
