What is in the Respiratory Bacterial Panel?

The TRUPCR® Respiratory Bacterial Pathogen Panel Kit is a real-time PCR assay designed to detect 14 clinically significant respiratory bacterial pathogens from a single respiratory sample. The test is structured across four multiplexed tubes, each using four fluorophores (FAM, HEX/VIC, ROX/Texas Red, Cy5) to identify multiple targets simultaneously.

About the TRUPCR® Respiratory Bacterial Pathogen Panel Kit

The kit is built on high-performance multiplex real-time PCR with:

  • Four pre-aliquoted multiplex tubes with validated primer/probe mixes
  • Hot-start DNA polymerase for enhanced specificity
  • Endogenous internal control for extraction and PCR quality checks
  • Compatibility with major qPCR instruments (ABI 7500 Fast, QuantStudio™ 5, Bio-Rad® CFX96)

Each target is designed against conserved genomic regions to ensure broad strain coverage and minimize cross-reactivity.

Principle and Workflow

  1. Sample Collection & Extraction
    Respiratory specimens such as BAL, tracheal aspirate, sputum, or nasopharyngeal aspirate in VTM are processed using standard total nucleic acid extraction methods.
  2. Multiplex PCR Setup
    The extracted nucleic acid is added to four separate tubes, each pre-loaded with a target-specific multiplex master mix containing primers and probes for designated bacterial pathogens.
  3. Real-Time Amplification
    The assay runs for 40 cycles on a compatible Real-Time PCR instrument, with fluorescence detection across four channels: FAM, HEX/VIC, ROX, and Cy5.
  4. Result Interpretation
    The endogenous internal control confirms sample quality and assay validity. Positive controls generate expected Ct values, supporting reliable interpretation of detected respiratory bacterial targets.
