Principle and Workflow
- Sample Collection & Extraction
Respiratory specimens such as BAL, tracheal aspirate, sputum, or nasopharyngeal aspirate in VTM are processed using standard total nucleic acid extraction methods. - Multiplex PCR Setup
The extracted nucleic acid is added to four separate tubes, each pre-loaded with a target-specific multiplex master mix containing primers and probes for designated bacterial pathogens. - Real-Time Amplification
The assay runs for 40 cycles on a compatible Real-Time PCR instrument, with fluorescence detection across four channels: FAM, HEX/VIC, ROX, and Cy5. - Result Interpretation
The endogenous internal control confirms sample quality and assay validity. Positive controls generate expected Ct values, supporting reliable interpretation of detected respiratory bacterial targets.
