Ultra Deep Microbiome Prep 10 Kit

Ultra-Deep Microbiome Prep10 isolates bacterial and fungal DNA from body fluids of 1 to 10 ml and from tissue pieces of about 0.5 by 0.5 cm, after depleting at least 95 per cent of the host DNA. Research use only. The chemistry is the same as in Ultra-Deep Microbiome Prep; the difference is the sample volume. The larger volume raises the number of microbial cells that reach the column, which matters when the load is low.

Ultra-Deep Microbiome Prep10 isolates bacterial and fungal DNA from larger sample volumes, 1 to 10 ml, with the same host depletion as the 1 ml version.

Principle

A chaotropic buffer lyses the human cells in the sample while microbial cells stay intact. The human DNA released, including DNA from dead cells, is degraded by the chaotrope-resistant DNase MolDNase B. The microbial cells are then collected and their cell walls degraded with BugLysis, followed by Proteinase K digestion and spin-column purification of the microbial DNA. The microbial cells are concentrated by centrifugation before lysis, which is what allows the larger input volume to be used. The manufacturer states removal of at least 95 per cent of host DNA, and lysis has been shown on over 200 bacterial genera and 65 fungal genera.

Sample material and equipment

Body fluids: ascites, bronchoalveolar lavage, blood, cerebrospinal fluid, pleural fluid, pus and synovial fluid, 1 to 10 ml. Tissue pieces of about 0.5 by 0.5 cm. Besides a thermomixer and a bench-top microcentrifuge, a high-speed centrifuge for 50 ml tubes at 9,500 x g is required. For volumes up to 1 ml the manufacturer offers Ultra-Deep Microbiome Prep.

What is and is not supplied

Buffers and consumables at room temperature, enzymes frozen, and 50 ml centrifuge tubes as a third component. No controls are supplied; an internal PCR control is available separately as Ultra-Deep Microbiome Ampli IC (G-130-042). Shelf life is 24 months.
