HLA-A3101 PCR Assay

Short description

The Viennalab RealFast™ Assays for the HLA-A31:01 allele provide a reliable and efficient way to prevent severe hypersensitivity reactions in patients prescribed carbamazepine. Carbamazepine can cause severe hypersensitivity reactions such as Stevens-Johnson syndrome (SJS), toxic epidermal necrolysis (TEN), and severe drug reaction with eosinophilia and systemic symptoms (DRESS) in approximately 5% to 10% of individuals.

The HLA-A31:01 allele is associated with a wider range of carbamazepine-induced hypersensitivity in diverse populations. Genotyping patients prior to starting therapy can significantly reduce the incidence of severe hypersensitivity reactions.

HLA-A3101 RealFast™ PCR Kit for Carbamazepine Hypersensitivity Screening

Carbamazepine is a widely prescribed medication for conditions such as epilepsy, bipolar disorder, and nerve pain. However, for some patients, it can cause severe hypersensitivity reactions like Stevens-Johnson syndrome (SJS), toxic epidermal necrolysis (TEN), and drug reactions with eosinophilia and systemic symptoms (DRESS). These conditions are potentially life-threatening, especially in individuals carrying certain genetic markers. The HLA-A31:01 and HLA-B15:02 RealFast™ PCR kits from Viennalab offer a quick and reliable way to screen for these genetic variants, helping clinicians identify patients who are at higher risk before starting carbamazepine therapy.

The HLA-A31:01 allele is associated with a wider range of hypersensitivity reactions across various populations, including Europeans and Japanese. By using the RealFast™ assays to screen for these alleles, healthcare providers can significantly reduce the risk of severe adverse reactions, as recommended by the FDA and the Clinical Pharmacogenetic Implementation Consortium (CPIC).

This PCR kit provides fast and accurate results, making it easier to make informed decisions about whether to prescribe carbamazepine. Both assays are simple to use and highly reliable, ensuring that patients receive the safest possible treatment. With the HLA-A31:01 RealFast™ PCR kits, you can offer personalized care and minimize the risk of serious drug reactions in your patients.

Components of the HLA-A3101 PCR Assay
  • DNA Extraction Kit: A kit containing all the reagents necessary for the extraction of genomic DNA from a blood or saliva sample, including DNA extraction buffer, proteinase K, and magnetic beads.
  • PCR Mix: A ready-to-use mix containing all the reagents necessary for the PCR amplification of the HLA-A and HLA-B gene regions of interest.
  • HLA-A3101 Primer Mix: A mix of primers specific for the HLA-A*3101 allele.
  • Probe Mix: A mix of probes specific for the detection of the HLA-A3101 allele.
  • Control DNA: A synthetic DNA that is added to the sample as an internal control to ensure that the assay is working correctly.
  • DNase/RNase-free Water: Water that has been treated to remove contaminating DNases and RNases, which can degrade the extracted DNA.

The kit also includes instructions for use, as well as a protocol for the detection of the HLA-A3101 allele using real-time PCR.
