Integrating automated extraction ahead of PCR

Abstract

How the SwiftXtractor SL fits in front of common Real-Time PCR platforms, automated nucleic-acid extraction, lower hands-on time, and cleaner input for downstream amplification.

Method & sample types

The SwiftXtractor™ SL (Xpedite) automates nucleic-acid extraction ahead of PCR using a reverse-purification chemistry, taking manual pipetting and timing variability out of what is often the least standardised step in a molecular workflow. It accepts common clinical sample types, the extraction chemistry itself is sample-type agnostic in the sense that it is designed around nucleic-acid capture rather than a specific matrix, so the same instrument supports multiple downstream assay types run in the same lab.

Controls & interpretation

Extraction quality is not something a downstream PCR run can fully compensate for: poor yield, retained inhibitors or degraded nucleic acid all show up as a weak or absent internal-control signal on whichever assay is run afterwards. Standardising the extraction step with an automated platform reduces one whole category of “why did this run fail” troubleshooting, since the pre-analytical variable that is hardest to control manually, consistency, is handled by the instrument instead of the operator.

Workflow integration

Positioned in front of standard Real-Time PCR platforms, the SwiftXtractor SL reduces hands-on time per batch and lets a single operator manage extraction for multiple downstream assays in parallel rather than extracting manually for each. For labs scaling up sample throughput without scaling headcount proportionally, automated extraction is typically the highest-leverage single change to a PCR workflow.
