LAMP for malaria: a 30-minute isothermal answer

How the Ampliflash Malaria LAMP kit detects Plasmodium without thermal cycling, a fast, low-instrument route to a result, and where it complements rather than replaces microscopy.

Method & sample types

Loop-mediated isothermal amplification (LAMP) amplifies target DNA at a single, constant temperature, which removes the need for a thermal cycler entirely. The BioSynex Ampliflash® Malaria LAMP kit uses this chemistry to detect Plasmodium directly from blood within a 30-minute workflow, from sample to result, using comparatively simple instrumentation.

Controls & interpretation

An internal amplification control is included to confirm the reaction proceeded normally, independent of the Plasmodium-specific result, important given that LAMP’s visual or fluorescent read-out formats rely on a clean, unambiguous positive/negative call rather than a quantitative curve. As with any rapid molecular malaria test, a positive result should prompt confirmatory species identification and parasitaemia quantification by microscopy where clinical management depends on it (e.g. distinguishing P. falciparum from non-falciparum species, or assessing severity).

Workflow integration

The isothermal format and short turnaround make LAMP a strong fit for settings without ready access to a thermal cycler or a skilled microscopist on every shift, it does not replace microscopy where speciation and parasite density matter clinically, but it substantially shortens time-to-first-answer for ruling malaria in or out, which is often the most time-critical decision in a febrile returning traveller or endemic-area patient.
